Peptide Reconstitution Math: Concentration and Volume

Reconstitution math relates the peptide mass in a vial to the final volume of solution. It can calculate concentration and the volume containing an already specified dose.

Start with two known values

You need the total peptide mass in the vial, in mg, and the final solution volume, in mL. Vial capacity is not the same as final solution volume. For example, a container that can hold 3 mL does not necessarily contain 3 mL of solution.

Calculate concentration

Concentration (mg/mL) = vial mass (mg) ÷ final solution volume (mL).

A hypothetical 10 mg vial in 2 mL of final solution has a concentration of 5 mg/mL. The same mass in 4 mL would have a concentration of 2.5 mg/mL. Increasing the volume lowers the concentration; it does not increase the peptide mass.

Convert a specified amount to liquid volume

Volume (mL) = specified amount (mg) ÷ concentration (mg/mL).

At 5 mg/mL, an already specified amount of 0.25 mg corresponds to 0.05 mL. Check the result by multiplying: 0.05 mL × 5 mg/mL = 0.25 mg. If the amount is in micrograms, convert mcg to mg first.

Added water and final volume

The calculator assumes that the water volume entered equals the final solution volume. This approximation requires any volume change from dissolving the material to be negligible. Use the final volume specified in validated product instructions; the calculator cannot verify that assumption.

What the math does not tell you

This guide does not choose a diluent, mixing volume, dose, storage method, or route of administration. It also cannot confirm identity, purity, complete dissolution, or uniform concentration. Follow product-specific instructions from a qualified professional.
